Description The Verhoeff Elastic Masson Trichrome Stain offers a comprehensive method for simultaneously staining elastic, muscle, and connective tissue, making it ideal for routine use in histopathology. Verhoeff’s technique renders the elastica black, creating a distinct contrast with the red-stained muscle and green or blue-stained connective tissue, achieved through a modification of Masson’s trichrome. Additionally, cell nuclei exhibit reddish brown to blue-black hue with Weigert’s iron hematoxylin. This kit is particularly well-suited for examining vascular pathology in experimental pathology, surgical and autopsy sections. Kit Components Sku# Reagent Name Volume (ml) VB-3034-1 Ethanol saturated with picric acid 250 ml VB-3034-2 Verhoeff’s elastic stain solution A 150 ml VB-3034-3 Verhoeff’s elastic stain solution B 60 ml VB-3034-4 Verhoeff’s elastic stain solution C 60 ml VB-3034-5 Biebrich scarlet-acid fuchsin solution 250 ml VB-3034-6 Phosphotungstic/phosphomolybdic Acid Solution 250 ml VB-3034-7 Aniline Blue Solution 250 ml VB-3034-8 1 % acetic acid solution 250 ml Storage Store at room temperature. Protocol Deparaffinize in xylene (6 minutes) ×2 Rehydrate: Ethanol 100% (2 minutes) ×2; Ethanol 95% (2 minutes) ×2; Ethanol 70% (2 minutes) ×1. Rinse in distilled water (2 minutes) ×1. Mordant in ethanol saturated with picric acid for 5 minutes Wash sections in tap water for 5 min until yellow color disappears. Freshly prepare modified Verhoeff’s elastic stain solution: mix Verhoeff’s elastic stain solution A, B and C at a ratio of 2.5:1:1. Stain in modified Verhoeff’s elastic stain for 6 min. Wash in running tap water for 10 min. Rinse in distilled water for 1 min. Stain in Biebrich scarlet-acid fuchsin solution for 2-3 min Place in Phosphotungstic/phosphomolybdic Acid Solution for 15 min. discard solution. Transfer directly into Aniline Blue Solution for 5 minutes. 1% Acetic acid for 2-3 seconds discard solution. Dehydrate with 2 changes of 95% Ethanol and 2 changes of 100% Ethanol (2 minute per change). Clear with 2 changes of xylene (5 minute per change). Mount cover slip onto glass slide with Permount or some other suitable organic mounting medium.
